Collection: Dopaminergic mechanisms underlying normal variation in trait anxiety
Description: Resting-state functional connectivity with bilateral amygdala as seed region (defined from each participant's freesurfer (v5.1) segmentation. Voxel-wise regression with amygdala dopamine release as a predictor. We measured dopamine release using a 2 PET scan protocol. To measure spontaneous dopamine release, we collected two [11C]raclopride PET scans per participant. We contrasted baseline [11C]raclopride D2/3 receptor binding and D2/3 binding following oral methylphenidate administration. Endogenous dopamine release was calculated as the percent change in BPND from scan 1 to scan 2 ((placebo [11C]raclopride – methylphenidate [11C]raclopride)/placebo [11C]raclopride). Methylphenidate blocks the dopamine transporter, which increases extracellular dopamine and leads to reduced [11C]raclopride D2/3 receptor binding via competitive displacement. The regression map here reflects the negative association between functional connectivity and dopamine release.
